Comparison of isozyme transformation in maize as a result of insertion of the chitinase gene Phyton
Yan,PM; Zhang,HF; Wang,Q; Yan,XY; Sun,Y.
Isozymes of peroxidase (POD), catalase (CAT), esterase (EST) and superoxide dismutase (SOD) were analyzed on transgenic maize (with external chitinase gene) and its parent by vertical polyacrylamide gel electrophoresis (PAGE). This study was made using shoots at the fourth leaf stage. Results showed that: POD and EST were detected in 6 bands. POD-2 and POD-3 were present at the bud and seedling stages. POD-1, POD-4, POD-5 and POD-6 were only present at the seedling stage. POD-6 expressed stronger in the transgenic maize with chitinase than in its parent. EST-2 was present only at the bud stage, and its expression in transgenic maize was stronger than that in its parent. EST-5 only existed at the seedling stage. EST-4 did not exist in the parent maize...

Diversity and genetic structure in natural populations of Hancornia speciosa var. speciosa Gomes in northeastern Brazil Rev. Bras. Frutic.
Martins,Georgia Vilela; Martins,Luiza Suely Semen; Veasey,Elizabeth Ann; Lederman,Ildo Eliezer; Silva,Edson Ferreira da.
Hancornia speciosa Gomes is a fruit tree native from Brazil that belongs to Apocinaceae family, and is popularly known as Mangabeira. Its fruits are widely consumed raw or processed as fruit jam, juices and ice creams, which have made it a target of intense exploitation. The extractive activities and intense human activity on the environment of natural occurrence of H. speciosa has caused genetic erosion in the species and little is known about the ecology or genetic structure of natural populations. The objective of this research was the evaluation of the genetic diversity and genetic structure of H. speciosa var. speciosa. The genetic variability was assessed using 11 allozyme loci with a sample of 164 individuals distributed in six natural populations...

Genetic stability in rice micropropagation Biocell
Medina,R; Faloci,M; Marassi,M.A.; Mroginski,L.A..
An efficient clonal propagation procedure for six rice varieties cultivated in Argentina was developed by using shoot tip cultures, and the genetic stability of the micropropagated plants was verified by isozyme analysis. One week old seedlings obtained on MS medium were sectioned and subcultured on MS medium (0.75% agar) supplemented with different combination and concentrations of cytokinins (BAP and KIN) and auxins (2,4-D and NAA). After four weeks of culture, multiple shoots were obtained. The best response was observed on MS supplemented with BAP 5 mg l-1. Shoot clumps were multiplied in MS liquid medium containing BAP 5 mg l-1. Profuse rooting was obtained after transfer to MS medium lacking growth regulators and with sucrose 8% (w/v). Complete...

Tambaqui (Colossoma macropomum) is among the most important fish species of the Amazon and one of the most cultivated in Brazil. In the present work we have evaluated the genetic variability of wild and captivity populations of C. macropomum. Enzymatic markers were used to estimate the genetic variability of 41 specimens from a wild group; and 30, 33 and 45 from three captivity groups, which came from Pentecostes (Ceará State), Jaboticabal (São Paulo State) and Itacoatiara (Amazonas State), respectively. Nine isoenzymic systems were used to evaluate the genetic variability of these populations. Using zimogram data we obtained the polymorphism level, allele number, allelic frequency, observed and expected heterozigosity, Wright F statistics (FIS, FST),...

Variability among inbred lines and RFLP mapping of sunflower isozymes Genet. Mol. Biol.
Carrera,Alicia D.; Pizarro,G.; Poverene,M.; Feingold,S.; León,A.J.; Berry,S.T..
Eight isozyme systems were used in this study: acid phosphatase (ACP), alcohol dehydrogenase (ADH), esterase (EST), glutamate dehydrogenase (GDH), malate dehydrogenase (MDH), phosphoglucoisomerase (PGI), 6-phosphogluconate dehydrogenase (PGD), and phosphoglucomutase (PGM). The polymorphism of these enzyme systems was studied in 25 elite inbred lines. A total of 19 loci were identified, but only eight of them were polymorphic in the germplasm tested. The polymorphic index for the eight informative markers ranged from 0.08 to 0.57, with a mean value of 0.36. Five isozyme loci were mapped in F2:3 populations with existing RFLP data. Est-1, Gdh-2 and Pgi-2 were mapped to linkage groups 3, 14 and 9, respectively. As in previous reports, an ACP locus and a PGD...
